Split-DNAzyme cooperating primer exchange reaction for sensitive miRNA detection

Abstract Sensitive quantification of microRNA (miRNA) plays a crucial role in early diagnosis and precise therapy osteosarcoma. Herein, we build label-free sensitive miRNA approach based on the activation split-DNAzyme initiated primer exchange reaction (PER). Target cooperates with Mg 2+ through assisting assembly DNAzyme to correct conformation, which enables performance PER-based nucleic acids amplification produce large amount single-strand DNA (ssDNA) sequences. The G-quadruplexes (G4) ssDNA sequences products bind N -methyl mesoporphyrin IX (NMM) form G4-NMM complex enhanced fluorescence respond. results demonstrate that miRNA-21 can assist split-DNAzyme, active exhibits high specificity efficiency inducing subsequent PER. Based split-DNAzyme-assisted signal recycle PER, method eventually shows sensitivity selectivity, providing promising prospect for stage tumor more therapy.
